A lot of the RAM that Vista uses is for caching applications it thinks you are likely to launch soon, so it can load them much quicker when (if) you do launch them. All the RAM used for this purpose will be released to applications if required, so the total RAM shown as available in Task Manager is not really a true representation of how much is actually available.
