Yes, my motherboard supports 6Gb/s

So are the speeds really that different and noticeable? Or Should I go stick with 3.0 even tho it has a lower cache

Buy the Samsung. Mechanical hard drives don't even come close to saturating SATA 3, let alone 6. You would not notice a difference at all. The Samsung is also a very quick drive, faster than the WD Black's in many cases.
